1. To stand up. Primarily heard in US. After turning off the alarm, I got on up and made my way over the dresser to fetch a shirt and trousers. Come on, get on up—I didn't hit you that hard!
2. To climb or ascend something. Primarily heard in US. We'd better get on up that peak before morning breaks and it gets too hot. Let me get on up the ladder and see how the roof is holding up.
